Discover how combining the power of BioLegend’s LEGENDplex™ multiplex immunoassays with the advanced Omni Bead Ruptor Elite™ (BRE) homogenizer can revolutionize tissue biomarker quantification. Our solutions allow you to move beyond traditional serum and plasma biomarkers to uncover local immune responses in tissues, opening doors to groundbreaking scientific insights.
While serum and plasma biomarkers provide valuable systemic insights, they often miss the localized immune dynamics occurring within tissues. The LEGENDplex system enables precise quantification of up to 14 cytokines and biomarkers in complex biological samples, including tissue homogenates. When paired with the Omni Bead Ruptor Elite, researchers can efficiently lyse even the toughest tissue samples, ensuring high-quality analyte recovery for downstream analysis.
Proper tissue lysis and complete biomarker release are fundamental to generating accurate, reproducible downstream data. When tissue lysis is inadequate, you face incomplete analyte release, leaving analytes trapped within cells, or overly harsh conditions that may degrade analytes. Even if your current homogenization method works, it may not deliver the efficiency and reproducibility your research demands. Unoptimized sample preparation doesn't just waste valuable time and resources—it puts your data quality at risk, potentially compromising the validity of your findings and delaying critical insights. Level-up your tissue lysis with the Omni Bead Ruptor Elite paired with optimized Pre-Filled Lysing Kits.
In our latest webinar, we utilized an LPS-induced mouse model of cytokine release syndrome to quantify key inflammatory markers from tough tissue samples like skin. Mouse skin was homogenized with either standard sonication methods or lysed using the Omni Bead Ruptor Elite before using the LEGENDplex Mouse Cytokine Release Syndrome (13-plex) Panel to perform cytokine quantification.

Overview of experimental workflow comparing cytokine quantification in skin processed using sonication or lysed with the Omni Bead Ruptor Elite.
Several cytokines, including CXCL10 (IP-10), CCL3 (MIP-1a), and IL-6 were detected at higher concentrations when processed with the Bead Ruptor Elite compared to samples processed using sonication, highlighting how optimized tissue lysis methods are essential for accurate and reliable cytokine detection.

Omni Bead Ruptor Elite (BRE) detects higher levels of cytokines in skin compared to sonication (Sonic). Concentrations shown are normalized per mg of total protein.
