Whole cell extracts (15 µg total protein per lane) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-STAT4 (clone W22042B). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405306). Direct-Blot™ HRP anti-GAPDH (Cat. No. 405405) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
IHC staining with Purified anti-STAT4 (clone W22042B) was performed on formalin-fixed paraffin embedded human testis tissue following antigen retrieval using Tris-EDTA pH9.0 Antigen Retrieval Buffer (Cat. No. 422704). The tissue sections were incubated without (panel A) or with Purified anti-STAT4 (clone W22042B) (panels B and C) followed by Alexa Fluor® 647 goat anti-rat (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801) (panels A and C). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 20X objective. Scale bar: 100 µm

Signal Transducer and Activator of Transcription 4 (STAT4) is a critical transcription factor involved in mediating immune responses. STAT4 is primarily activated via phosphorylation which leads to its dimerization, nuclear translocation, and subsequent activation of target genes involved in immune regulation. STAT4 plays a pivotal role in the differentiation of T helper 1 (Th1) cells, follicular helper T cells (Tfh), and regulatory T cells (Tregs), and is essential for the function of natural killer (NK) cells and CD8+ T cells. STAT4 expression is tightly regulated and varies across cell types and activation states, with higher expression observed in activated immune cells such as mature dendritic cells and monocytes. Genetic polymorphisms in the STAT4 gene, such as H623Y and A635V, have been linked to altered STAT4 activity and nuclear localization, influencing susceptibility to various diseases, such as rheumatoid arthritis, systemic lupus erythematosus, type 1 diabetes, and inflammatory bowel disease.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
