Interleukin-17A (IL-17A), originally identified as cytotoxic T lymphocyte-associated antigen-8 (CTLA-8) in mice, is a founding member of the IL-17 cytokine family. IL-17A is primarily produced by CD4+ Th17 cells, but also by CD8+ T cells, γδ T cells, and innate lymphoid cells. It signals through a receptor complex of IL-17RA and IL-17RC, activating NF-κB and MAPK pathways, which drive the expression of cytokines, chemokines, and antimicrobial peptides including IL-6, IL-8, G-CSF, GM-CSF, GRO-α, and MCP-1. It plays a key role in the pathogenesis of autoimmune diseases, and IL-17A-targeted therapies are being developed for chronic inflammatory disorders.
BioLegend’s ELISpot MAX™ Deluxe Set includes pre-optimized essential reagents, along with additional buffers and solutions, for conducting an Enzyme-Linked Immunosorbent Spot (ELISpot) assay. This kit is specifically designed to detect human IL-17A-secreting cells. An antibody specific for human IL-17A is coated onto an ethanol-activated 96-well PVDF membrane plate as the capture. Stimulated and/or unstimulated cells are pipetted directly into the wells. The plate is incubated at 37°C for a designated period, during which the capture antibody binds to IL-17A from the secreting cells in the vicinity. After incubation, the cells are removed, and a biotinylated anti-human IL-17A detection antibody is added, followed by avidin-HRP. Next, the TMB membrane substrate is added. The TMB substrate reacts with HRP to form a blue-black precipitate at the sites where IL-17A is present. The result is the appearance of visible spots on the membrane surface, representing the footprints of individual IL-17A-secreting cells.
