Glutathione peroxidase 4 (GPX4) is a selenoenzyme that plays a critical role in protecting cells from oxidative damage by reducing phospholipid hydroperoxides within membranes, thereby preventing lipid peroxidation and ferroptosis. GPX4 is essential for maintaining membrane integrity and redox homeostasis, particularly in tissues rich in polyunsaturated fatty acids like the brain and kidneys. Its expression and activity are tightly regulated through transcriptional, post-transcriptional, epigenetic, and proteostatic mechanisms, and depend on selenium availability and glutathione synthesis. Dysregulation or deficiency of GPX4 is implicated in a range of diseases, including cancer (especially therapy-resistant head and neck cancers), neurodegeneration, infertility, acute kidney injury, and Sedaghatian-type spondylometaphyseal dysplasia.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 2.5 - 10.0 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
