Whole cell lysate from the indicated cell lysates (15 µg total protein) were resolved on a 4-12% Bis-Tris gel, transferred to a PVDF membrane, and probed with Purified anti-SIX3 antibody (clone W23154C). Proteins were visualized by chemiluminescence detection using HRP goat anti-rat IgG antibody (Cat. No. 405405). Direct-Blot™ HRP anti-β-actin Antibody (Cat. No. 643807) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker -
Jurkat cells (negative cell line, panel A) and WERI-Rb-1 cells (positive cell line, panel B) were fixed with Fixation Buffer (Cat. No. 420801), permeabilized with 0.5% Triton X-100, and then blocked with 5% FBS. Cells were then stained with Purified anti-SIX3 (clone W23154C) followed by Alexa Fluor® 647 Goat anti-rat IgG antibody (Cat. No. 405416). Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective. Scale bar: 50 µm -

The SIX3 protein is a transcription factor that plays a crucial role in the development of the forebrain and eyes. It functions by binding to specific DNA sequences, regulating the expression of target genes involved in these developmental processes through both repression and activation. During human forebrain development, SIX3 represses WNT1 expression to ensure proper patterning. Mutations in the SIX3 gene are associated with holoprosencephaly type 2 (HPE2), a severe congenital brain malformation characterized by incomplete separation of the forebrain. Additionally, SIX3 mutations have been linked to schizencephaly, a rare brain disorder that results in abnormal clefts or slits in the cerebral hemispheres.

ICC, ICFC - Verified - Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using either True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401).
For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using PFA-Methanol or PFA-Triton X-100. We do not recommend Methanol only fixation/permeabilization - RRID
