SMAD5 (mothers against decapentaplegic homolog 5) is a receptor-regulated SMAD protein primarily involved in the bone morphogenetic protein (BMP) signaling pathway, which is part of the broader TGF-β superfamily. Upon BMP receptor activation, SMAD5 is phosphorylated and forms a complex with SMAD4, translocating to the nucleus to regulate gene transcription related to cell differentiation, proliferation, apoptosis, and tissue homeostasis. Beyond its canonical role, non-phosphorylated SMAD5 also influences energy metabolism by promoting mitochondrial respiration and glycolysis in response to cytoplasmic pH changes. Its activity is tightly regulated through phosphorylation, nuclear-cytoplasmic shuttling, and interactions with other proteins such as SMURF1, which mediates its degradation. Dysregulation of SMAD5 has been implicated in several diseases, particularly respiratory disorders such as COPD, asthma, pulmonary arterial hypertension, idiopathic pulmonary fibrosis, and lung cancer.

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
