HAP1 wild-type cells (positive cell line, top panels) (HAP1 parental cell line, Horizon Cat No. C631) and HAP1 XIAP Knockout cells (negative cell line, bottom panels) (HAP1 XIAP KO cell line , Horizon Cat No. HZGHC008092c010) were fixed with Fixation Buffer (Cat. No. 420801), permeabilized with ice-cold methanol, and blocked with 5% FBS for 1 hour at room temperature. Cells were then stained with Purified anti-XIAP (clone W23268E), followed by incubation with Alexa Fluor® 647 Goat anti-rat IgG (Cat. No. 405416) for 1 hour at room temperature. Nuclei were counterstained with DAPI (Cat. No. 422801). The images were captured on Revvity Operetta CLS™ High Content Analysis System with a 40X objective and merged. Scale bar: 50 µm

Description

XIAP, also known as BIRC4, is one of the potent endogenous suppressors of apoptosis in humans. It belongs to the inhibitor of apoptosis protein (IAP) family, a group characterized by the presence of one or more baculoviral IAP repeat (BIR) domains that mediate interactions with apoptotic proteins. XIAP serves as a direct inhibitor of key effector caspases, functioning as a crucial regulator of programmed cell death and cellular homeostasis.

XIAP inhibits the proteolytic activity of caspase-3, caspase-7, and caspase-9, the principal executioners of the intrinsic apoptotic pathway. The BIR2 domain and the adjacent linker region inhibit caspase-3 and caspase-7, while the BIR3 domain inhibits caspase-9. Blocking XIAP prevents the downstream cleavage of cellular substrates that culminates in apoptotic disassembly. XIAP also contains a RING finger E3 ubiquitin ligase domain, allowing it to ubiquitinate caspases and other target proteins, which can direct them toward proteasomal degradation.

Overexpression of XIAP is observed in many cancers, including colorectal, pancreatic, breast, and hematological malignancies. Mutations in the XIAP/BIRC4 gene impair its regulatory functions, especially in pathways involving NOD2 signaling and immune homeostasis. Patients with XIAP deficiency exhibit heightened susceptibility to hemophagocytic lymphohistiocytosis (HLH), severe inflammatory responses, chronic colitis resembling Crohn’s disease, and dysregulated immune cell survival.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25- 5.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is 5 µL per 106 cells in 100 µL volume. It is highly recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).

For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following:
-Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100
-Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol
-100% ice-cold methanol only

For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with either Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803), True-Phos™ Perm Buffer (Cat. No. 425401) or True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401).
