Recombinant Human PDGFRα-Fc Chimera (carrier-free) (Cat. No. 569906) (black circles) inhibits the proliferation of NIH3T3 mouse embryonic fibroblast cells in the presence of Recombinant Human PDGF-AA (carrier-free) (Cat. No. 773706). Ultra-LEAF™ purified anti-human PDGFRα (clone W22075L) (purple squares) inhibits the effect of Recombinant Human PDGFRα-Fc Chimera (6 µg/mL) in a dose-dependent manner, whereas the Ultra-LEAF™ Purified Rat IgG2b, κ Isotype Control Antibody (Cat. No. 400644) (gray triangles) does not have the effect. ND50 range: 1.5 – 10 µg/mL.

The Platelet-derived growth factor (PDGF) family comprises of four different polypeptide chains encoded by four genes. The four PDGF chains assemble into disulfide-bonded dimers via homo- or heterodimerization. Five different dimeric isoforms have been described so far; PDGF-AA, PDGF-AB, PDGF-BB, PDGF-CC, and PDGF-DD. PDGF is a major mitogen for fibroblasts, smooth muscle cells, and others. They exert their functions by interacting with two related receptor tyrosine kinases, PDGF receptor α (PDGFRα) and PDGF receptor β (PDGFRβ). Upon ligand binding, PDGFRα undergoes dimerization and autophosphorylation, resulting in the formation of docking sites for SH2 domain-containing signaling proteins. These recruited molecules include enzymatic effectors (e.g., Src family kinases, phosphatidylinositol 3-kinase, Phospholipase Cy1, rasGAP, tyrosine phosphatase Syp) and adaptor proteins (e.g., GRB-2, Shc, Nck). The SH2 domains of these proteins specifically recognize phosphorylated PDGFRα, mediating the activation of downstream signaling pathways. PDGFRα plays a key role in many diseases through the above signal transduction.
