Human CD3+ T cells were isolated from healthy human PBMCs using the MojoSort™ Human CD3 T Cell Isolation Kit (Cat. No. 480022). Isolated cells were left untreated or either expanded in vitro with BioLegend’s Human CD3/CD28/CD137 T Cell Activation Beads or BioLegend’s Human CD3/CD28 T Cell Activation Beads (Cat. No. 422603) at 1:1 particle-to-cell ratio in the absence of human IL-2. On day 7 post-activation with the beads, the number of expanded cells was quantified using AOPI staining. Cell counts for each condition were expressed as a ratio relative to the untreated control. -
On day 7 after activation with Human CD3/CD28/CD137 T Cell Activation Beads, the expanded T cells were co-stained with PE/Cyanine7 anti-human CD4 (Cat. No. 300511) and Brilliant Violet 510™anti-human CD8a (Cat. No. 301047). Dead cells were excluded by Helix NP™ Blue (Cat. No. 425305) staining. The CD4+ and CD8+ T cells were analyzed by flow cytometry (left panel), and the percentage of CD4-/CD8a+ T cells in the expanded samples was quantified (right panel). -
On day 7 after activation with Human CD3/CD28/CD137 T Cell Activation Beads, the expanded T cells were stained for flow cytometric analysis. To select for Central Memory T cells (CD8+CCR7+CD45RA-), the cells were initially gated on CD4-CD8+ expression. Then, cells were analyzed with PE/Fire™ 640 anti-human CD197 (CCR7) (Cat. No. 353261) and Spark Red™ 718 anti-human CD45RA (Cat. No. 304183) (left panel). The proportion of Central Memory CD8+ T cells (CD8+CCR7+CD45RA-) was determined (right panel). As a comparison, T cells were also activated with BioLegend’s Human CD3/CD28 T Cell Activation Beads (Cat. No. 422603).

The Human CD3/CD28/CD137 T Cell Activation Beads allow for convenient in vitro activation of human T cells without the need for antigen-presenting cells or precoating culture plates. Human T cells are stimulated with microbeads coated with monoclonal anti-CD3, anti-CD28 and anti-CD137 antibodies. The antibodies present on the bead surface act as an agonist and stimulate T cells. This mimics antigen presenting cells to induce the proliferation, especially for memory T cells which rely on CD137 signaling for cell maintenance and activation. Activation in vitro with this product results in robust T cell expansion, especially of the central memory (CCR7+ CD45RA-) CD8+ T cell subset.
Simply add the beads directly to isolated T cells at a 1:1 particle-to-cell ratio for robust T cell activation. The antibody-coated beads can be removed from the cells using a magnetic separator such as the MojoSort™ Magnet (Cat. Nos. 480019, 480020, and 480173). Some of the downstream applications include functional assays, gene expression analysis, and phenotypic characterization.
