CD11b, encoded by the integrin alpha M (ITGAM) gene, pairs with CD18 (integrin β2) to form the heterodimeric receptor Mac-1 or complement receptor 3 (CR3). This receptor is primarily expressed on myeloid cells, including neutrophils, monocytes, macrophages, dendritic cells, and some natural killer cells. In these cells, CD11b mediates adhesion, migration, and phagocytosis. Functionally, CD11b/CD18 binds to several ligands such as iC3b, ICAM1, and fibrinogen, allowing leukocytes to adhere to endothelium and clear opsonized pathogens. It also plays an important role in immune regulation, suppressing excessive inflammation by inhibiting TLR signaling and promoting tolerance in certain contexts. Genetic variants in ITGAM have been associated with autoimmune diseases, including systemic lupus erythematosus, highlighting CD11b function in maintaining immune homeostasis.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.1 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.

For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
