Whole cell extracts (15 µg total protein) from indicated cell lines were resolved by 4-12% Bis-Tris gel electrophoresis, transferred to a PVDF membrane, and probed with Purified anti-mouse p53 (clone W22179B). Proteins were visualized by chemiluminescence detection using HRP Goat anti-rat IgG (Cat. No. 405405). Direct-Blot™ HRP anti-GAPDH (Cat. No. 607904) was used as a loading control. Western-Ready™ ECL Substrate Premium Kit (Cat. No. 426319) was used as a detection agent. Lane M: Molecular weight marker

The p53 protein plays a crucial role as a tumor suppressor by regulating the cell cycle, promoting DNA repair, and inducing apoptosis in response to cellular stressors such as DNA damage. Its function is tightly regulated by proteins like Mdm2 and Mdm4, which inhibit p53 activity under normal conditions and prevent unnecessary cell death. In clinical research, mouse models have been invaluable for understanding p53 role in cancer biology. These models help elucidate how p53 mutations contribute to tumor development and progression, providing insights that are directly translatable to human cancers. The study of p53 in mice has also facilitated the development of targeted therapies aimed at reactivating p53 function in tumors, highlighting its importance in advancing cancer treatment.
