Description

XRCC1 (X-ray repair cross-complementing protein 1) is a key scaffold protein involved in the repair of DNA single-strand breaks (SSBs). It orchestrates the assembly of DNA repair complexes by interacting with key enzymes such as DNA ligase III, DNA polymerase β, and poly (ADP-ribose) polymerase 1 (PARP1). XRCC1 is recruited to DNA damage sites via its BRCT1 domain, which binds to poly (ADP-ribose) (PAR) chains synthesized by activated PARP1, while its BRCT2 domain facilitates interactions with other repair proteins, including PARP2. Regulation of XRCC1 function is finely tuned by both DNA damage-induced and constitutive protein interactions. For instance, components of the SAGA complex, such as GCN5 and TADA2B, bind XRCC1 independently of DNA damage and modulate its recruitment and retention at damage sites, revealing a dual regulatory mechanism that can become dysfunctional in disease-associated XRCC1 variants. Mutations or deficiencies in XRCC1 are linked to several human diseases, including Spinocerebellar Ataxia Autosomal Recessive 26 (SCAR26) and Xeroderma Pigmentosum Group D, and are associated with increased cancer susceptibility due to impaired DNA repair capacity.

Recommended Usage

Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 10 μg/mL is recommended. For flow cytometric staining using our True-Phos™ Perm Buffer, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401).

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
