Description

Calpain-1 is a calcium-dependent cysteine protease that plays a role in modulating cellular architecture and signaling. Calpain-1 has a heterodimeric structure consisting of two distinct subunits that work together to form the active enzyme. It cleaves various substrates involved in cytoskeletal remodeling, cell motility, and apoptosis. Its activity is tightly regulated by intracellular calcium levels and by its endogenous inhibitor, calpastatin, which ensures precise control to prevent unwanted proteolysis. Dysregulation of Calpain-1 has been implicated in several diseases, including neurodegenerative disorders like Alzheimer's and Parkinson's, cardiovascular diseases, and cancer, where altered proteolytic activity contributes to pathological cell migration, inflammation, and tissue damage.

Recommended Usage

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 1.25 - 10.0 μg/mL is recommended. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

Additional Product Notes

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401).

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
