The ZO-2 protein is a key component of tight junctions. It plays a role in maintaining cell polarity and barrier function. ZO-2 interacts with other tight junction proteins like claudins and occludins and participates in signaling pathways that influence gene expression and cell proliferation. Its expression is regulated by cellular stress, cytokines, and developmental cues. Mutations in TJP2 have been associated with several diseases, including progressive familial intrahepatic cholestasis type 4 (PFIC4), familial hypercholanemia, and autosomal dominant non-syndromic hearing loss (DFNA51).

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.0 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
For use of this antibody in Immunocytochemistry (ICC), we recommendf ixation and permeabilization using PFA-Triton X-100 or PFA-ice-cold methanol. We do not recommend ice-cold methanol only.
