Interleukin 10 (IL-10), also known as CSIF, GVHDS, is a cytokine which plays a critical role in protecting the host from excessive responses to pathogens, while also participating in wound healing, cancer, and homeostasis. Through its binding to the IL-10 receptor, a complex of IL-10R1 and IL-10R2, IL-10 functions as an anti-inflammatory cytokine by down-regulating TH-1 cytokine and promoting development of regulatory T cells (Treg). Major sources of IL-10 include T helper cells, monocytes, macrophages, and dendritic cells. However, other cells such as B cells and epithelial cells are also capable of producing IL-10. Abnormally low level of IL-10 can cause autoimmunity and chronic inflammation (e.g. colitis), while high level of IL-10 can result in immune deficiency and tumor tolerance.
BioLegend’s ELISpot MAX™ Deluxe Set contains pre-optimized essential reagents and additional buffers and solution for Enzyme-Linked Immunosorbent Spot assay. This kit is designed for detection of human IL-10 secreting cells. A monoclonal antibody specific for human IL-10 is coated on ethanol-activated 96-well PVDF membrane plate as the capture. Stimulated and/or unstimulated cells are pipetted directly into the wells, and then incubated at 37°C for a specific period of time. During this period, the capture antibody binds to human IL-10 from the secreting cells in the vicinity. Then, cells are removed and a biotinylated anti-human IL-10 detection antibody is added, followed by Avidin-HRP. Subsequently, TMB membrane substrate is added. The TMB membrane substrate reacts with HRP to form a blue-black precipitate at the sites of human IL-10 secreting cells. The end result is visible spots, which corresponds to footprints of IL-10 secreting cells, on the membrane surface.
