CD112R is a cell surface receptor that competes with CD226 to bind to CD112 (Nectin-2). CD112R-CD112 interaction inhibits T cell proliferation, whereas disruption of this interaction enhances T cell responses. These findings support its role as a co-inhibitory receptor, or immune checkpoint molecule, that suppresses TCR-mediated signaling.
Structurally, CD112R is a single-pass transmembrane protein with one extracellular IgV domain. Its long intracellular domain contains two tyrosine residues, including one within an ITIM-like motif, consistent with inhibitory signaling capacity. CD112R was initially named PVRIG based on homology between its second exon and the variable immunoglobulin domain of the poliovirus receptor (PVR/CD155) and poliovirus receptor-like (PVRL) gene family.

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is 5 µL per million cells in 100 µL staining volume or 5 µL per 100 µL of whole blood. It is recommended that the reagent be titrated for optimal performance for each application.
