HeLa (top panels) and A549 cells (bottom panels) were fixed with Fixation Buffer (Cat. No. 420801), permeabilized with 0.5% Triton X-100, and blocked with 5% FBS. Cells were then stained with Purified anti-SUMO1 (clone 66AT1273.94) (middle and right panels) or Purified Mouse IgG1, κ Isotype Control antibody (left panels) (Cat No. 400102), followed by incubation with Alexa Fluor® 647 Goat anti-mouse IgG (Cat. No. 405322). Nuclei were counterstained with DAPI (Cat. No. 422801) and co-stained with Alexa Fluor® 488 anti-tubulin-α (Cat. No. 627906) (merged in left and middle panels). The images were captured on a Revvity Operetta CLS™ High Content Analysis System with a 63X objective and merged. Scale bar: 50 µm -
HAP1 WT cells (positive cell line, filled histogram) (Horizon Cat. No. C631) and HAP1 SUMO1 Knockout cells (negative cell line, open histogram) (Horizon Cat. No. ZGHC002501c006) were fixed and permeabilized using True-Phos™ Perm Buffer (Cat. No. 425401) and intracellularly stained with Purified anti-SUMO1 (clone 66AT1273.94) or Purified Mouse IgG1, κ Isotype Control (dashed histogram, representative of both cell lines) (Cat No. 400102), followed by Alexa Fluor® 647 Goat anti-Mouse IgG (Cat. No. 405322).

SUMO1 (Small Ubiquitin-like Modifier 1) is a protein modifier that regulates key cellular processes such as transcription, DNA repair, and protein stability through a reversible process called sumoylation. SUMO1 attaches to target proteins via a cascade involving E1, E2, and E3 enzymes, and is removed by SUMO-specific proteases. SUMO1 also influences protein interactions and localization such as nuclear transport of target proteins. Its regulation includes enzymatic control and autoinhibitory mechanisms that affect substrate specificity and stress responses. Dysregulation of SUMO1 is linked to several diseases, including cleft lip and palate, heart failure, cancer, and neurodegenerative disorders like Alzheimer’s disease, highlighting its importance in development and cellular homeostasis.

Each lot of this antibody is quality control tested by formalin-fixed paraffin-embedded immunohistochemical staining. For immunohistochemistry, a concentration range of 2.5 - 10.0 µg/mL is suggested. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.

For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401).
For use of this antibody in immunocytochemistry (ICC), we recommend fixation/permeabilization with the following combinations:
-PFA and ice-cold methanol
-PFA and Triton X-100
For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval with Citrate Buffer (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
