CD234, also known as atypical chemokine receptor 1 (ACKR1), controls chemokine levels and localization via high-affinity chemokine binding. Unlike classical chemokine receptors, ACKR1 does not initiate ligand-induced signal transduction, but instead mediates chemokine sequestration, degradation, or transcytosis. ACKR1 interacts with inflammatory chemokines of the CC and CXC subfamilies, but not with homeostatic chemokines. Known ligands include CCL2, CCL5, CCL7, CCL11, CCL13, CCL14, CCL17, CXCL5, CXCL6, CXCL8 (IL-8), CXCL11, GRO, RANTES, MCP-1, and TARC.
ACKR1 regulates chemokine bioavailability and, consequently, leukocyte recruitment via two distinct mechanisms. In endothelial cells, it mediates the abluminal to luminal transcytosis of tissue-derived chemokines, enabling their activity on circulating leukocytes. In erythrocytes, ACKR1 functions as both a reservoir and a sink for circulating chemokines, buffering surges in plasma chemokine levels. Additionally, ACKR1 serves as a receptor for the malaria parasite Plasmodium yoelii in mature erythrocytes. The N-terminal glycosylated extracellular domain of ACKR1 carries the Duffy blood group antigens Fya and Fyb.

Each lot of this antibody is quality control tested by immunofluorescent staining with flow cytometric analysis. For flow cytometric staining, the suggested use of this reagent is ≤ 0.5 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
